Evidence for a Novel Countertransport Mechanism*

چکیده
Adenine transport in the L1210 leukemia cell is mediated by a rapid facilitated process. Evidence supporting a carrier transport mechanism includes the observations that 1) extracellular hypoxanthine or xanthine inhibits net adenine uptake and 2) the presence of these bases within the intracellular compartment produces an uphill flow of adenine into the cell (countertransport). On the other hand, a very interesting phenomenon is observed when ATP-depleted cells are exposed simultaneously to extracellular adenosine and C3H]adenine in that there is a sustained uphill flow of C3H]adenine into the cell. A similar uphill flow of C3H]adenine into the cell results during simultaneous exposure of cells to inosine and I3HIadenine. Examination of the transport and metabolism of adenosine and its metabolites by high performance liquid chromatography demonstrates the basis for this phenomenon. Adenosine enters the cell by a mechanism distinct from that for adenine and is rapidly converted to inosine which can be further metabolized to hypoxanthine. Both of these compounds can flow out of the cell down their concentration gradients. The uphill flow of 13H]adenine into the cell is a result of a countertransport phenomenon with hypoxanthine in which hypoxanthine competes with intracellular j3H]adenine for efflux by the adenine carrier. Consistent with this hypothesis are the effects of deoxycoformycin and persantin. Persantin, an inhibitor of nucleoside transport, abolishes adenosine or inosine stimulation of adenine uptake by blocking the entry of these nucleosides into the cell. Deoxycoformycin, an inhibitor of adenosine deaminase, blocks only the adenosine stimulatory effect by preventing formation of inosine and consequently the subsequent conversion to hypoxanthine. As expected, deoxycoformycin does not alter the stimulatory effect of inosine on uphill transport of adenine.

Studies finding an increased maximal rate of Na-Li countertransport in red blood cells from persons with essential hypertension and their normotensive offspring have raised the possibility that Na-Li countertransport may serve as a marker for the genetic predisposition to hypertension. We studied Na-Li countertransport in 238 randomly selected blood donors representative of the population of Ro...

Abnormalities in erythrocyte Li-Na countertransport have been reported in hypertensive subjects, and the available evidence favors familial aggregation and striking heritability of this marker. It is uncertain, however, whether the abnormalities are associated with hypertension per se or whether they may be concentrated in a particular subset of hypertensive subjects. In the present study, maxi...

Several laboratories have reported that the activities of sodium-lithium countertransport are increased in red blood cells from patients with essential hypertension. Based on the many similarities between this transport system and the renal sodium-proton exchanger, a hypothesis has been put forth in the literature that increased red blood cell sodium-lithium countertransport activity may be a m...

Both Na+/Li+ countertransport and electrochemical proton gradient (delta mu(H+))-induced Na+ and H+ fluxes are increased in erythrocytes from patients with essential hypertension. It was assumed that these abnormalities are related to ubiquitous (housekeeping) forms of the Na+/H+ exchanger (NHE-1). To examine this hypothesis, we compared kinetic and regulatory properties of erythrocyte Na+/Li+ ...
